EHA 2026 | Findings from MonumenTAL-3: Tal+Dara±Pom versus Dara+Pom+Dex in R/R multiple myeloma

Peter Voorhees, MD, Levine Cancer Institute, Charlotte, NC, discusses the key findings from the pre-planned interim analysis of the MonumenTAL-3 trial (NCT05455320) comparing talquetamab (Tal) in combination with daratumumab (Dara), with or without pomalidomide (Pom), to Dara with Pom and Dexamethasone (Dex) in patients with relapsed/refractory (R/R) multiple myeloma. Dr Voorhees highlights that the efficacy of Tal+Dara±Pom was encouraging and that the regimen resulted in significant improvements in progression-free survival (PFS) and overall survival (OS) compared with the control arm. He also notes the distinct side effect profile of the regimen due to on-target, off-tumor effects of Tal; however, he emphasizes that taste changes, skin and nail changes, and cerebellar side effects were generally manageable. This interview took place at the 31st Congress of the European Hematology Association (EHA) in Stockholm, Sweden.
