Stéphane de Botton, MD, PhD, Gustave Roussy Institute, Paris, France, comments on the results and molecular characterization of patients from the AGILE study (NCT03173248), which investigated ivosidenib plus azacitidine versus placebo plus azacitidine in patients with newly diagnosed IDH1-mutated acute myeloid leukemia (AML). Dr de Botton highlights the frequency and clearance of mutations observed in patients, including IDH1, NRAS, DNMT3A, TET2, and ASXL1, and also comments on the emergence of new mutations. Overall, the study demonstrated that ivosidenib plus azacitidine leads to deep and durable responses, showed significant clinical benefit compared to placebo, and achieved clearance of all baseline mutations in this difficult-to-treat population.
