ASCO 2026 | Could BTKis play a role in DLBCL through rational combinations?

In this video, Manali Kamdar, MD, University of Colorado Cancer Center, Denver, CO, briefly discusses whether BTK inhibitors (BTKis) may still play a role in diffuse large B-cell lymphoma (DLBCL) through rational combinations, highlighting the promising data with acalabrutinib, lenalidomide, and rituximab in the upfront setting. This interview took place during the 2026 American Society of Clinical Oncology (ASCO) Meeting in Chicago, IL.

So the data that we have with acalabrutinib-lenalidomide-rituximab in the upfront setting is very intriguing, seems very promising. You don’t really lose a lot of bang for the buck if patients are not in remission and you add chemotherapy towards the back end. So I do believe that there is immense interest in developing BTK inhibitors in DLBCL. It’s all about precision medicine, targeting it for the right subtype at the right time.
