Rakesh Popat, BSc, MBBS, MRCP, FRCPath, PhD, University College London Hospitals NHS Foundation Trust, London, UK, explores the potential of cereblon E3 ligase modulatory drugs (CELMoDs), such as iberdomide and mezigdomide, as an alternative to traditional immunomodulatory agents in the treatment of relapsed/refractory (R/R) multiple myeloma (MM). CELMoDs show activity in those patients who are refractory to both lenalidomide and pomalidomide, with mezigdomide demonstrating encouraging efficacy in higher-risk patients.
