ASCO 2022 | MagnetisMM-3: elranatamab in relapsed/refractory multiple myeloma

Lesokhin, MD, Memorial Sloan Kettering Cancer Center, New York City, NY, discusses safety results from the MagnetisMM-3 trial (NCT04649359), a Phase II, open-labeled, multicenter, non-randomized trial evaluating elranatamab, a B-cell maturation antigen (BCMA)-CD3 bispecific antibody in patients with relapsed/refractory multiple myeloma. Patients enrolled were refractory to at least 1 proteasome inhibitor, 1 immunomodulatory drug, and 1 anti-CD38 antibody. Preliminary results of MagnetisMM-3 in patients with relapsed/refractory myeloma and no prior BCMA-targeted treatment suggest that 76 mg of elranatamb administered once weekly with a 2-step-up priming regimen is well tolerated, with no grade 3 cytokine release syndrome or immune effector cell-associated neurotoxicity syndrome (ICANS) observed.
