New approaches to target TP53-mutated AML: the ADAPT trial

Andrew Wei, MBBS, PhD, Peter MacCallum Cancer Centre & Royal Melbourne Hospital, Melbourne, Australia, discusses new approaches to target TP53-mutated acute myeloid leukemia (AML), highlighting the potential role of TP53 inactivation and subsequent BCL-xL upregulation in treatment failure after venetoclax-based therapy. Dr Wei explains the rationale and dosing schedule of the ALLG AMLM28/A2 (ADAPT) trial (ACTRN12625000994482p), a Phase Ib/II study evaluating the addition of pelcitoclax (APG-1252) to AML therapy for patients with TP53 aberrations or persistent measurable residual disease (MRD). This presentation took place at the 8th International Workshop on Acute Leukemias (iwAL) in Cambridge, MA.
