ERIC CLL 2026 | How is frontline fixed-duration acalabrutinib plus venetoclax shaping treatment decisions in CLL?

Alexey Danilov, MD, PhD, City of Hope, Duarte, CA, discusses the impact of the AMPLIFY trial (NCT03836261) on his practice, noting that fixed-duration acalabrutinib combined with venetoclax has become a standard frontline option for most patients with previously untreated chronic lymphocytic leukemia (CLL) due to its high efficacy and safety profile. Prof. Danilov notes that any newer combination, such as sonrotoclax plus zanubrutinib, would need to demonstrate at least non-inferiority, and potentially improved progression-free survival and overall efficacy, to displace acalabrutinib-venetoclax. This interview took place at the 2026 ERIC International Meeting in Barcelona, Spain.
