SOHO 2026 | Insights into the potential of nuvisertib plus momelotinib in patients with myelofibrosis and anemia

The momelotinib-nuvisertib combination actually is a very intriguing one in the sense that, you know, it’s actually the first. So in that sense, it’s a trendsetter. It’s the first to explore a momelotinib backbone for a rational synergistic combination study in myelofibrosis. Now, there is also a study of momelotinib with luspatercept called ODYSSEY, which I led, but that one, you know, is more looking to augment anemia responses, whereas this one is really a PIM kinase, PIM1 kinase plus JAK kinase inhibitor combination...

The momelotinib-nuvisertib combination actually is a very intriguing one in the sense that, you know, it’s actually the first. So in that sense, it’s a trendsetter. It’s the first to explore a momelotinib backbone for a rational synergistic combination study in myelofibrosis. Now, there is also a study of momelotinib with luspatercept called ODYSSEY, which I led, but that one, you know, is more looking to augment anemia responses, whereas this one is really a PIM kinase, PIM1 kinase plus JAK kinase inhibitor combination. And so a synergistic mechanism-based combination, and the first one to use a momelotinib backbone. So, you know, we are expecting data on quite a few more patients at this year’s ASH, but so far, you know, what has been shown, this is at actually last year’s ASH, is data on 18 patients from the momelotinib plus nuvisertib combination. And now there was also an updated EHA of 2026. So I think what is important to note, though, is that the initial 18 patient data that were presented last year at ASH were patients who took the combination in the fasted state. So now the development has moved to the fed state. So I think it’s important to distinguish between these two sets of results. So again, you know, we are seeing very promising, you know, spleen symptom and anemia responses to the combination. And I want to make a point about the anemia. Obviously, anemia is already momelotinib strength, but it was also recently found that nuvisertib also may improve anemia by blocking ACVR1 and down-regulating hepcidin. So two drugs doing the same thing in that regard. And then spleen symptoms as well. So it’s been very promising. We’ll see hopefully even more patients reported on at ASH. And again, you know, if you compare historically to the MOMENTUM data, which is really the design that this study is sort of based on, you see that momelotinib plus nuvisertib is giving you quite a bit better spleen and symptom responses than momelotinib alone. So I think MOMENTUM, obviously this is not a randomized trial at this point, but MOMENTUM serves as a good comparison because that is what this is based on.
