CAR-T Meeting 2026 | The safety and efficacy of var-cel (ARI-0001) in patients with R/R mantle cell lymphoma

Nil Albiol, MD, Hospital Clínic de Barcelona, Barcelona, Spain, discusses the findings of a study evaluating the safety and efficacy of varnimcabtagene autoleucel (var-cel; ARI-0001) in patients with relapsed/refractory (R/R) mantle cell lymphoma (MCL). Dr Albiol notes that the results were encouraging, with the main factor influencing overall survival (OS) being whether or not the patient achieved a complete response (CR). This interview took place at the EBMT-EHA 8th European CAR T-cell Meeting, held in Palma de Mallorca, Spain.

In this poster we reported the results of var-cel in mantle cell lymphoma. We included 30 patients, but 27 did eventually reach CAR-T infusion. So from these 27 patients, 90% had achieved some kind of response, 78% a complete response, and the median duration of response was 17 months with a median PFS of 15 months. Now we have follow-up of around 22 months and median OS is around three years, which is quite remarkable...

Now we have follow-up of around 22 months and median OS is around three years, which is quite remarkable. And the main factor influencing overall survival was whether or not the patient achieved a complete response. So for those patients who achieved the CR, the median OS was not reached versus seven months for those who did not get a CR. And regarding safety, var-cel had a very favorable safety profile with 93% of patients had some kind of CRS, but most of them were grade one. And we had only one single case of ICANS and CRS grade 3 or higher which occurred in the same patient and it was a patient with a high disease burden and some other complications.
