Combination strategies in AML: triplets, targeted therapies and venetoclax resistance

Hima Atluri, MD, Northwestern University, Chicago, IL, shares insight into current and emerging combination strategies in acute myeloid leukemia (AML), including and beyond the azacitidine and venetoclax backbone. Dr Atluri highlights when upfront triplets or targeted therapy may be more beneficial in certain molecular subgroups, such as ivosidenib-based triplets in IDH1-mutated AML, and how venetoclax resistance may be overcome by switching to cladribine-based regimens, particularly in monocytic and RAS-mutated AML. She also discusses targeting surface antigens with pivekimab sunirine, including in TP53-mutated AML, and outlines results from the Phase Ib/II study (NCT04086264). This presentation took place at the 8th International Workshop on Acute Leukemias (iwAL) in Cambridge, MA.
