Novel targets for T-cell engagers in relapsed/refractory AML

David Sallman, MD, Moffitt Cancer Center, Tampa, FL, presents evidence for targeted antigen immunotherapy approaches in acute myeloid leukemia (AML), including data from the Phase I study (NCT05143996) of the FLT3xCD3 bispecific T-cell engager CLN-049 in patients with relapsed/refractory (R/R) AML and myelodysplastic syndromes (MDS). Dr Sallman highlights that CLN-049 was well tolerated, with durable responses in heavily pretreated patients, and mentions the upcoming Phase Ib study (NCT07722767) of CLN-049 with azacitidine and venetoclax in the frontline setting. Finally, he discusses other emerging immunotherapeutic approaches, including T-cell engagers targeting cathepsin G, CD70-directed CAR T-cells secreting a T-cell engager, and novel combination strategies. This presentation took place at the 8th International Workshop on Acute Leukemias (iwAL) in Cambridge, MA.
