Alexander Lesokhin, MD, Memorial Sloan-Kettering Cancer Center, New York, NY, discusses current and emerging targets for multiple myeloma monoclonal antibodies (MAbs). Novel bispecific antibody treatments such as elranatamab, which binds to CD3 on T-cells and B-cell maturation antigen (BCMA), have favorable efficacy and safety profiles. Cytokine release syndrome is a side effect of these treatments, but is manageable with steroids and IL-6 antagonists such as tocilizumab. Bispecific antibodies with alternative targets highly expressed on multiple myeloma cells such as GPRC5D and FcRH5 have also shown promise.
